Zyagen offers a full range of immunohistochemistry (IHC) services, including routine immunohistochemistry (peroxidase and alkaline phosphatase), immunofluorescence (IF), immunocytochemistry (ICC), apoptosis detection (TUNEL), and proliferation assays (BrdU).
IHC can be done on paraffin and frozen tissue sections of any tissue type of any species. We can use ready to stain slides provided by customers or by Zyagen. We can also process experimental tissues provided by customers for paraffin or frozen OCT embedding, sectioning, and immunostaining.
For accuracy of each IHC, negative and positive control sides will be run in parallel with the experimental tissue samples.

Immunohistochemistry Validation Services
For successful and consistent immunohistochemical staining, it is critical to optimize the experimental conditions of each antibody to enhance detection and reduce background. Optimization includes
- Optimization of tissue fixation including selection of suitable fixative agent and determine the optimal incubation time with the fixative.
- Optimization of antigen retrieval to expose antigen to the antibody by detergent, enzymes or boiling with sodium citrate EDTA, and Tris-EDTA buffers with and without pressure.
- Titration of the primary antibody by testing at different concentrations and incubation times.
- The optimal adjusted conditions of the antibody will be used for testing the experimental samples.

Routine Immunohistochemistry and Immunofluorescence Services
- Processing tissue samples provided by clients or by Zyagen for paraffin or OCT frozen embedding, sectioning, and making slides ready-to-use for immunostaining.
- Tissue samples weather paraffin sections, frozen sections, cultured cells, or floating sections are immunostained with selected well validated antibody that is specific to antigen of your interest in tissues of any species. Antibodies can be provided by clients or purchased from reliable commercial sources
- Detection method of immunostaining is up to the client weather enzymatically (Peroxidase or Alkaline Phosphatase immunostaining) or by fluorescent staining (Immunofluorescence).
- Interpretation of immunostaining results by our expert scientists.

Double Immunohistochemistry Services
We also perform immunofluorescence on paraffin or frozen tissue sections to localize at least two proteins/antigens simultaneously on the same tissue sections.
- The well-established validated primary antibodies are tested on samples weather paraffin sections, frozen sections, cultured cells, or floating sections.
- Detection is by fluorescent staining (Immunofluorescence) with different colors.
- As a counterstaining, we are routinely using DAPI.
- Interpretation of immunostaining results by our expert scientists.
- Sending high quality microscope digital images with report of immunostaining. Four Images will be taken for each immunostaining, DAPI, first antibody, second antibody, and one for all of them (merge images).

TUNEL Assay (Apoptosis Detection Assay)
Zyagen offer TUNEL staining for the detection of apoptosis in paraffin sections, frozen sections, cell suspensions, and cell cultures.
- DNA will be labeled by terminal deoxynucleotidyl transferase (TdT) and detected by enzyme end labeling (peroxidase) or fluorescent labeling.

BrdU Assay (Proliferation Detection Assay)
Bromodeoxyuridine (5-bromo-2′-deoxyuridine, BrdU) is a synthetic thymidine nucleoside analog. BrdU is commonly used to study proliferation of living cells in vivo and in vitro. Dividing cells incorporate BrdU into newly synthesized DNA during the S-phase of the cell cycle and the BrdU can then be detected with anti-BrdU specific antibodies by flow cytometry, immunofluorescence, and immunohistochemistry. BrdU has been also extensively used to study cellular proliferation in rodent where it has been administered by ip injection.
- Processing cultured cells incubated with BrdU or tissues harvested from rats or mice injected with BrdU for paraffin or OCT embedding, and sectioning.
- Immunostaining Paraffin or frozen sections with specific antibody to BrdU.
- Detection of immunostaining by enzymatic (Peroxidase) or fluorescent staining (Immunofluorescent)
