of blots. Protein samples are extracted from tissues provided by Zyagen or
customers. To ensure high quality blots, each protein sample whether provide by the
customers or Zyagen
is subjected to rigorous quality control test before use. We can make blots with up to
22 tissues
each plus the protein marker.
Procedures of construction of Western
blots:
Harvesting fresh tissues.
Homogenization of tissues in RIPA lysis buffer supplemented with SDS
and a cocktail of 7 mammalian protease inhibitors.
Extraction of tissue lysates and quantitation of total
protein using sensitive and accurate assay.
Testing the purity and integrity of protein by gel electrophoresis.
Fractionation of protein (75 microgram) by
large SDS-denatured
polyacrylamide gel
electrophoresis.
Transfer protein onto PVDF membranes.
Shipping blots at room temperature in sealed bags.
