Rat Hippocampus/thalam/hypothalam Coronal Floating Frozen Sections Reg.8. Frozen Floating Sections are specially made for immunostaining in culture plates or tubes (not on slides). Brains are fixed in 10% neutral buffered formalin (NBF), cryoprotected in serial agarose solutions, snapped frozen in OCT blocks and sectioned at a thickness of 30 microns. Sections are placed in very cold 12-well culture plates.

Quality Control: Tissue sections are H&E stained and examined by histologists with 25 years of experience to be sure of excellent morphology and high quality.

Applications:
Frozen floating sections are prepared in RNase-, DNase-, and protease-free environment and are ideal for immunohistochemistry and in situ hybridization.

Packing/shipping: Frozen floating sections are shipped in 12-well culture plates on dry ice and are good for one year if properly stored.

Handling sections: The frozen thick sections are shipped in culture plates (one section per each well) on dry ice. Upon arrival, immediately keep culture plates at -80oC and never allow them to warm-up to the room temperature (sections will attach to the culture plates at RT). For transferring sections to another plates or tubes, always use very cold forceps (sections will attach to warm forceps)

Prior to use: Take plates off the freezer and immediately place them on flat piece dry ice. Gently add 1-2 ml PBS to the bottom of each well (not on the sections) to allow sections floating in the buffer. Gently shake plate by hand to flatten the sections in PBS. It is recommended to re-fix the sections in 10% neutral buffered formalin for 10 min before immunostaining. In this case, add 1-2 ml fixative to each well instead of the PBS. After addition of PBS or fixative, remove plates from dry ice and follow the immunostaining protocol of floating sections at room temperature.

If you want to use only small number of sections, transfer sections to a new dry ice-cold culture plate using a dry ice-cold forceps. Restore the leftover sections in the culture plates in the deep freezer.
